In Blue Eye Samurai, the 12 and 20 positions are sex positions, as mentioned by Madame Kaji, who runs a tea house near the harbor.

Mizu and Ringo arrive at Madame Kaji’s tea house and Mizu insists on meeting with her.

After a long wait, Madame Kaji appears before Mizu. She tells Mizu that any woman can study woodcuts and master the 12 and 20 positions.

She continues and states that to master the way of battle, a samurai must learn every art, including sex.

Mizu tells Kaji that she is looking for Fowler, and Kaji is shocked because of Fowler’s brutal requests from the prostitutes.
